ABSTRACT

PURPOSE: Behcet's disease (BD) is a chronic, relapsing, multi-system vasculitis of unknown aetiology with complicated and diversified clinical features predominantly involving oral and genital ulcers, and ocular and cutaneous lesions. The clinical features of this disease have been described to be different according to geographical areas and gender. We investigated the specific clinical features of BD patients in Northeastern China. MATERIALS AND METHODS: 116 patients involved in this study fulfilled the classification criteria of the International Study Group for BD. The clinical manifestations and results of laboratory tests of BD were recorded in each patient. RESULTS: The onset was typically between 20-39 years with a slight female predominance. Oral ulcers were the most common manifestation, followed by skin lesions, positive pathergy reaction/genital ulcers, and ocular lesions. Vascular lesion and epididymitis were rare in patients with BD. The frequency of erythema nodosum-like lesion and articular involvement were significantly higher in females, while gastrointestinal involvement was significantly higher in males. The results of laboratory tests showed that the human leukocyte antigen (HLA)-B*51 alleles were positive in 30.9% of patients and the immunological abnormities were present in some patients. CONCLUSION: The clinical features of BD showed geographical and gender difference. Genetic and immune factors might participate in aetiopathogenesis of BD.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate further the nature of HLA-DQB1 alleles association with dermatomyositis/polymyositis (DM/PM).</p><p><b>METHODS</b>HLA-DQB1 typing of 52 patients with DM/PM were investigated by polymerase chain reaction-sequence specific primer method, and the frequencies of HLA-DQB1 alleles were compared.</p><p><b>RESULTS</b>It was found that the patients with DM/PM had significantly increased frequency of HLA-DQB1*0401(RR=3.56, P=1.79 10(-3), Pc<0.05).</p><p><b>CONCLUSION</b>The results suggest that HLA-DQB1*0401 gene contribute to genetic susceptibility to DM/PM.</p>

ABSTRACT

To probe course and mechanism of cellular immune respone in early syphilitic lesion, the expression levels of CD3, CD20, CD68 were determined by immunohistochemistry and a computer image analysis system. The results showed that the expressions of CD3, CD20, and CD 68 were positive in 43 cases of primary and secondary syphilis. The expression level of CD68 was higher thanthat of CD3, CD20, especially around blood vessels. The expression level of CD 20 was very low. This study indicates that T cells, B cells, and in particular macrophages play an important cellular immunologic regulating role in clearance of Treponema pallidum from the infected tissue, but the efficacy is not high enough.

ABSTRACT

Objective:To explore the relationship between expressions of guococorticoid receptor(GR)? and ? mRNA in peripheral blood mononuclear cell(PBMC) of patients with polymyositis/dermatomyositis(PM/DM) and the glucocorticoid(GC) efficacy.Methods:Reverse transcription-polymerase chain reaction(RT-PCR) was used to semiquantite the expressions of GR? and ? mRNA in PBMC of 18 cases of PM/DM patients sensitive to GC,10 cases of PM/DM patients resistant to GC and 20 cases of health blood donors.The levels of cortisol in sera of PM/DM patients and of health donors were detected using radioimmunoassay.Results:There was no significant difference in the levels of seral cortisol among PM/DM patients sensitive to GC, resistant to GC and the health donors.The expressions of GR? mRNA in PBMC from PM/DM patients either sensitive to GC or resistant to GC were significantly lower those that of the normal control. Moreover,the expression of GR? mRNA in PBMC of PM/DM patients resistant to GC was significantly lower than that of patients sensitive to GC. While the expression of GR? mRNA in PBMC of PM/DM patients was significantly higher than that of the normal control,there was no significant difference of the expression of GR? mRNA between PM/DM patients sensitive and resistant to GC.Conclusion:The lower expression of GR? mRNA in PM/DM patients may be related to resistance to GC therapy,whereas there is no significant role of the expression level of GR? mRNA.

ABSTRACT

Objective:To investigate the relationship between pemphigus vulgaris(PV) and HLA-DR,DQ haplotypes in Han nations of northeast China.Methods:Polymerase chain reaction-sequence specific primers(PCR-SSP) method was used to detect the HLA-DRB1 and DQB1 alleles of 27 PV patients of Han nation of northeast China, analysed haplotyes and compared with 99 healthy controls.Results:Compared with control group, the frequencies of the haplotypes of HLA-DRB1*140x-DQB1*0503,DRB1*140x-DQB1*0201,DRB1*120x-DQB1*0503 and DRB1*140x-DQB1*0302 increased significantly in PV group. After statistical test, the difference between the two groups was significant.Conclusion:The special haplotypes may contribute to genetic susceptibility to PV in northeast Chinese.
